NKG2D (Natural Killer Group 2D) is a C-type lectin-like activating receptor encoded by the KLRK1 gene. It is expressed as a homodimer on the surface of NK cells, CD8⁺ T cells, γδ T cells, and others. The intracellular domain of NKG2D lacks signaling capability and requires the adaptor protein DAP10 to transmit activation signals, which activate the PI3K pathway to enhance immune cell cytotoxicity and cytokine release. NKG2D ligands are upregulated in over 80% of solid tumors and hematological malignancies (e.g., liver cancer, AML). Co-expression of multiple ligands across tumor subpopulations significantly reduces immune escape due to antigen loss. Senescent cells highly express MICA/MICB, enabling NKG2D CAR-T to selectively eliminate senescent cells and ameliorate age-related diseases.
Hillgene has developed an NKG2D CAR-T Premade lentivirus with specific killing functionality and convenient transduction efficiency evaluation. This virus utilizes the VSV-G envelope protein, enabling transduction of most mammalian cells, including primary and non-dividing cells. The virus employs a second-generation CAR structure design, incorporating the NKG2D ECD, CD8 hinge, 4-1BB costimulatory domain, and CD3ζ signaling domain. This design prolongs CAR-T cell survival and promotes proliferation. Additionally, the virus has been modified for Kanamycin resistance (KanR) to comply with domestic and international regulatory requirements.
This virus is produced using Hillgene's serum-free suspension packaging system and downstream lentiviral purification system (293T cell line), yielding a high-titer, low-impurity NKG2D CAR-T Premade lentivirus product. Customers can bypass complex lentiviral vector preparation for cell process optimization and validation while using this product to guide cell process development and accelerate workflows.
